11. Summary of the new findings of the thesis

- Two ORFs of OsDREB1A and OsDREB2A were successfully isolated  and sequenced from drought stress-treated cDNA library of Vietnam rice cultivar named Cuom Dang 1. The sequence analysis confirmes that both ORFs are identical with reference ones from japonica rice cultivar Nipponbare. Both ORFs were separately subcloned into pBIG-Ubi/Lip9 to create four expression vectors ready for A. tumefaciens-mediated transformation.

- By systematically screening callus induction and shoot generation ability of 47 Vietnamese rice cultivars, it was successful to estabilish the A. tumefaciens-mediatedtransformation protocol for Chanh trui variety.

- Four expression vectors: Ubi:OsDREB1A, Ubi:OsDREB2A, Lip9:OsDREB1A and Lip9:OsDREB2A were introducted into Chanh Trui variety, totally, 54 trangenic lines at T0 generation were confimed by PCR with two primer pairs.

- Finally, 04 trangenic lines (named L3 and L5 for Lip9:OsDREB1A; U1 and U4 for Ubi:OsDREB1A) at T3 generation were selected by genotyping and phenotyping assay. The transgenic plant could recuse and keep growing under dehydration condition.

12. Paratical applicability, if any:

The expression vectors containing the Vietnamese material can be supplied for improving the drought tolerance ability of other Vietnamese rice cultivars or other crops. Four transgenic lines can be used for breeding activities in order to created the drought tolerance rice varieties in Vietnam. In future, if GMO can be accepted to be free-existing in the field, the transgenic lines could be use directly in the agricultural system.

13. Further research directions, if any:

- Maintaining the stability genetics of transgenic lines in green-house condition.

- Study on transformation of expression vector in other crops likes maize, soybean, cotton…
